The Human IL-11 R alpha (Luc) HEK293 Reporter Cell was engineered to express STAT3 signaling response element. When stimulated with human IL-11 protein, the IL-11/ IL-11 R alpha interaction drives STAT3-mediated luminescence. Inhibition of IL-11 binding to IL-11 R alpha by either anti-IL-11 or anti-IL-11 R alpha antibodies results in a decrease in luminescence.

Application
Inhibition of human IL-11 protein-induced reporter activity by anti-human IL-11 neutralizing antibody.
This reporter cell was incubated with serial dilutions of antibodies in the presence of human IL-11 protein (Cat. No. IL1-H5243) with a final concentration of 0.2 μg/mL. The EC50 of anti-human IL-11 neutralizing antibody is approximately 1.047 μg/mL.

背景(Background)
IL-11 and IL-6 are signal transducer and activator of transcription 3 (STAT3) activators and frequently found upregulated in colorectal cancer (CRC). STAT3 activation in tumor epithelial cells is critically involved in the pathogenesis of CRC. Cancer-associated fibroblasts (CAFs) can modulate the tumor microenvironment in CRC. The latest research indicated that phosphorylation of STAT3 (pSTAT3) expression in CAFs is inversely correlated with the survival in CRC patients. IL-6 and IL-11 induce partly overlapping gene expression patterns in fibroblast subsets in CRC. In a summary, STAT3 activation in CAFs plays a key role in the cross-talk to the tumor epithelium.
