描述(Description)
The Human 5-HT1A (Luc) HEK293 Reporter Cell was engineered to not only express CREB signaling response element, but also express the receptor full length human 5-HT1A (Gene ID: 3350) and mGFP by a viral P2A self-cleaving peptide. When stimulated with human 5-HT1A agonist, the agonist/5-HT1A interaction drives CREB-mediated luminescence. Inhibition of activated 5-HT1A signaling by corresponding inhibitors results in a decrease in luminescence.

Surface expression of human 5-HT1A on Human 5-HT1A (Luc) HEK293 Reporter Cell was confirmed by detection of mGFP (FITC) using flow cytometry.

Application
Response to human 5-HT1A agonist.
The Human 5-HT1A (Luc) HEK293 Reporter Cell was stimulated with serial dilutions of human 5-HT1A agonist (Serotonin). The EC50 of human 5-HT1A agonist was approximately 15.13 μM with the max induction fold 16.71.
Protocol
Inhibition of Serotonin-induced activity by human 5-HT1A antagonist.
This reporter cell was incubated with serial dilutions of human 5-HT1A antagonist in the presence of Serotonin with a final concentration of 10 μM. The EC50 of human 5-HT1A antagonist is approximately 0.0039 μM.

Passage stability analysis by Signaling Bioassay.
The continuously growing Human 5-HT1A (Luc) HEK293 Reporter Cell was stimulated with serial dilutions of human 5-HT1A agonist (Serotonin). Serotonin stimulated response demonstrates passage stabilization (fold induction and EC50) across passage 15-23.

背景(Background)
This gene encodes a G protein-coupled receptor for 5-hydroxytryptamine (serotonin), and belongs to the 5-hydroxytryptamine receptor subfamily. Serotonin has been implicated in a number of physiologic processes and pathologic conditions. Inactivation of this gene in mice results in behavior consistent with an increased anxiety and stress response. Mutation in the promoter of this gene has been associated with menstrual cycle-dependent periodic fevers.
